Abstract: | A cursorysearchoftheProteinDataBank(PDB)withthekeyword “de novo” returns 962entries,whileforthekeyword “designed”, 6225 entrieswerereturned[asofAugust2017].Althoughthesenumbers(de novo or designedstructures)are1000timeslesserin magnitude (comparedtothetotalnumberofstructuresthePDBholds,whichiscloseto135,000structures),these B7000 structures indicatehowfarthe field ofproteindesignhasadvanced,since1950s.Proteindesigningprojectsareambitiousintheir goal duetothesimpleyetcomplexproblemofproteinfolding.Muchhasbeenlearnedabouthowaproteinfolds,andthese fundamental knowledgehavehelpedfurthertheareaofproteindesigntoconceiveproteinswithimaginativestructures |
