Title: Production and Characterization of L-methionine Gamma Lyase from Different Bacterial Sources

Abstract: The aim of this study was to both validate production of and characterize methionine gamma lyase from three different bacterial sources that are selected through literature survey and in silico search. Production of MGL was observed in all three, with Pseudomonas lurida producing the enzyme only intracellularly whereas P. fragi and C. braakii produced the enzyme both extracellularly and intracellularly. The MGL production was found to be maximum at pH 6 and 30°C in case of P. lurida. Whereas P. fragi and C. braakii showed maximum MGL production at 35°C and pH 7. Enzyme characterization revealed highest enzyme activity at 40°C and pH 6 for P. lurida whereas enzyme activity was maximum at 40°C and pH 7 in case of P. fragi and C. braakii. The Km value of MGL from C. braakii was observed to be highest (0.88 mM for intracellular and 0.926 for extracellular), while that from P. lurida was observed to be 2.11 mM and from P. fragi 1.11 mM and 1.08 mM for extracellular and intracellular enzymes respectively.
